Pengembangan dan Validasi Metode Analisis Betametason dalam Tablet dengan Metode Absorbansi dan Luas Daerah di Bawah Kurva Secara Spektrofotometri Ultraviolet

Abstract

A precise, accurate, fast and economical method has been developed and validated for the analysis of betamethasone tablets using ultraviolet spectrophotometry with absorbance method and area under the curve. Determination of this method was carried out at maximum wavelength absorption of 241.40 nm and area under the curve in the range 210.40 - 290.40 nm using a 0.1 N hydrochloric acid solvent. The method of analysis was validated by linearity, detection limits, quantification limits, precision and accuracy. The linearity in absorption method shows the result of regression equation y = 0.02273 x + 0.00228 with r = 0.99958, with method of area under curve shows result of regression equation y = 0.643 x – 0.214 with r = 0.9955. Limit detection and quantification limits on absorbance method 1.754531 and 5.316812, with area wide method under curve 3.888631 and 11.783730. Accuracy obtained by the absorbance method and the area under the curve satisfy the validated requirements of 80-120%. The precision obtained by the absorbance method and the area under the curve satisfies the validated requirement with the RSD yield ≤ 2%. Statistical analysis showed that both methods did not differ significantly (P ≥ 0.05).
